چکیده

Degenerating chondrocytes are often localized in the lower hypertrophic and calcifying zone of rat epiphyseal cartilage. The ability of these degenerating chondrocytes to synthesize and secrete sulfated proteoglycans was investigated using the light microscopic radiosulfate labeling method and the high-iron diamine-thiocarbohydrazide-silver proteinate (HID-TCH-SP) method for sulfated complex carbohydrates at the ultrastructural level. At 30 min following intraperitoneal (IP) injection of 35SO4, only intracellular autoradiographic labeling was observed, whereas both intracellular and extracellular radiosulfate labeling was seen at 1 h postinjection. A giadual increase in intracellular and extracellular labeling was seen at 3, 12 and 24h following IP injection of 35SO4. In ultrastructural cytochemical preparations, HID-TCH-SP weakly to moderately stained the chondrocyte Golgi vacuoles or immature and intermediate secretory granules but did not stain Golgi saccules. Mature secretory granules and extracellular matrix granules demonstrated strong staining. These results therefore indicate that proteoglycans are sulfated in the degenerating chondrocyte Golgi vacuoles, packaged in the secretory granules and then released into the extracellular matrix to form matrix granules. Introduction Proteoglycans (PGs) are closely associated with calcification of cartilage [1-3]. PGs in cartilage exist as high-molecular-weight PG aggregates and their constituent subunits. PG monomer is composed of glycosaminoglycans (GAGs) covalently bound to core protein. GAGs found in cartilage are chondroitin sulfates A and C, keratan sulfate and hyaluronic acid. The latter acid is not covalently bound to core protein but plays a role in the linking of PG monomers [4]. Conditioned medium derived from mesenchymal Stem cells regenerates’ defected articular cartilage

Background & Aims: One of cell- based technical issues associated with cartilage repair assay is delivering cells to the site of the parts where damage is created. Mesenchymal stem cells (MSCs) with their chondrogenic potential are ideal candidates for cartilage regeneration. High expression of cartilage hypertrophy markers by MSCs would result in apoptosis and ossification. This investigation ...

Altered Synthesis of Cartilage-Specific Proteoglycans by Mutant Human Cartilage Oligomeric Matrix Protein

BACKGROUND The mechanism by which mutant cartilage oligomeric matrix protein (COMP) induces a pseudoachondroplasia phenotype remains unknown, and the reason why a mutation of a minor protein of the growth plate cartilage causes total disruption of endochondral bone formation has not yet been determined. The current study was performed to investigate the effects of mutated COMP on the synthesis ...

Study of Human Chondrocyte Redifferntiation Capacity in Three-Dimensional Hydrogel Culture

Objective(s) Articular cartilage tissue defects cannot be repaired by the proliferation of resident chondrocytes. Autologous chondrocyte transplantation (ACT) is a relatively new therapeutic approach to cover full thickness articular cartilage defects by in vitro grown chondrocytes from the joint of a patient. Therefore, we investigated the redifferentiation capability of human chondrocytes ma...

Quantitative Analysis of the Proliferation and Differentiation of Rat Articular Chondrocytes in Alginate 3D Culture

Background: While articular chondrocytes are among those appropriate candidates for cartilage regeneration, the cell dedifferentiation during monolayer culture has limited their application. Several investigations have indicated the usefulness of alginate, but the topic of proliferation and differentiation of chondrocytes in alginate culture has still remained controversial. Methods: Rat articu...

Co-Culture of Mesenchymal Stem Cells with Mature Chondrocytes: Producing Cartilage Construct for Application in Cartilage Regeneration

Background: Cell-based treatment approach using differentiated mesenchymal stem cells (MSCs) and mature chondrocytes has been considered as an advanced treatment for cartilage repair. We investigated the differentiated level of these two cell types that is crucial for their repair capacity for cartilage defect at a co-culture micro mass system. Methods: Passaged-2 MSCs isolated from the mouse b...
